The 9 Hallmarks of Aging: A Plain-English Guide

In 2013, scientists identified 9 biological processes that drive aging. Understanding them is the key to understanding every longevity intervention — from fasting to senolytics to NAD+ supplementation.

In 2013, a landmark paper in Cell identified 9 'hallmarks of aging' — the core biological processes that drive deterioration over time. Every major longevity intervention targets one or more of these hallmarks. Understanding them gives you a framework for evaluating any longevity claim.

1. Genomic instability: DNA damage accumulates over time. Your cells have repair mechanisms, but they become less efficient with age. Interventions: minimize radiation/toxin exposure, support DNA repair with NAD+ precursors.

2. Telomere attrition: Telomeres shorten with each cell division. Short telomeres trigger cellular senescence. Interventions: exercise, stress reduction, TA-65, Mediterranean diet.

3. Epigenetic alterations: Gene expression patterns change with age, often in harmful directions. Interventions: diet, exercise, and sleep can partially reverse epigenetic aging (Horvath clock).

4. Loss of proteostasis: Damaged proteins accumulate (amyloid, tau). Autophagy clears them. Interventions: fasting, spermidine, exercise all upregulate autophagy.

5. Deregulated nutrient sensing: mTOR, AMPK, and sirtuins — the longevity signaling pathways — become dysregulated. Interventions: caloric restriction, fasting, berberine, rapamycin, resveratrol.

6. Mitochondrial dysfunction: Mitochondria become fewer and less efficient. Interventions: Zone 2 exercise, NMN/NR, CoQ10, cold exposure.

7. Cellular senescence: Zombie cells accumulate and secrete inflammatory signals (SASP). Interventions: quercetin + dasatinib (senolytics), fasting.

8. Stem cell exhaustion: Tissue regeneration capacity declines. Interventions: exercise, fasting, and emerging stem cell therapies.

9. Altered intercellular communication: Inflammatory signaling increases (inflammaging). Interventions: anti-inflammatory diet, stress reduction, social connection.
